  本学位论文是在对冬青属植物化学成分和药理活性研究进展的综述基础上,对四季青的化学成分进行了研究。四季青为冬青科(Aquifoliaceae)冬青属(Ilex)植物冬青Ilex chinensis Sims的干燥叶,秋、冬二季采收叶,晒干,具有清热解毒,消肿祛瘀的功效,用于肺热咳嗽,咽喉肿痛,痢疾,胁痛,热淋;外治烧烫伤,皮肤溃疡。

  采用多种色谱分离方法和波谱解析技术,从冬青叶70 %乙醇提取物的乙酸乙酯萃取部位中分离并鉴定了42个化合物(1-42),其中新化合物11个(1-11)。分离得到的化合物包括34个三萜类化合物,分别为三萜苷元15个:四季青素A和B(1和2),乌苏酸(12),救必应酸-3,23-缩酮(13),2α,3β-二羟基-23-去甲-4(24)-12-二烯-乌苏酸(14),救必应酸(15),3β-羟基-27-p-(Z)-香豆酰基乌苏-12-烯-28-酸(16),23-羟基乌苏酸(17),3β,23-二羟基乌苏-12,18(19)-二烯-28-酸(18),2α,3β,23-三羟基乌苏-12,18-二烯-28-酸(19),积雪草酸(20),3β-羟基-27-p-(Z)-香豆酰基齐墩果-12-烯-28-酸(21),ilexosapogenin A(22),2α,3β-二羟基-23-去甲-4(24)-12(13)-二烯-齐墩果酸(23),29-羟基常春藤皂苷元(24);三萜皂苷19个:四季青苷A-I(3-11),长梗冬青苷(25),ilexoside B(26),坡模酸-28-O-β-D-葡萄糖酯苷(27),rotungenoside(28),3β,23-二羟基-12,18-二烯-乌索酸-28-O-β-D-葡萄糖酯苷(29),3β,23-二羟基-12,19-二烯-乌索酸-28-O-β-D-葡萄糖酯苷(30),3β,23-二羟基-12,19(29)-二烯-乌索酸-28-O-β-D-葡萄糖酯苷(31),ilexoside A(32),泰国树脂酸-28-O-β-D-葡萄糖酯苷(33),28-O-β-D-glucopyranoside spathodic acid(34);酚酸类化合物4个:香草酸(35),对羟基苯甲酸(36),4-羟基肉桂酸(37),原儿茶酸(38);甾体类2个:β-谷甾醇(39),胡萝卜苷(40);香豆素1个:七叶内酯(41);脂肪酸类1个:十六烷酸(42)。 化合物19,21,23,24为首次从该属中分离得到,除化合物12,15,25,26,32,38-40外,均为首次从该种植物中分离得到。

  在上述研究结果的基础上,对分离得到的化合物进行了体外抗炎、保肝、肿瘤细胞毒等活性筛选:化合物1,3,5,12,16,21,29,30在10-5 M浓度下,表现出较为显著的抑制小鼠巨噬细胞NO生成的活性;化合物4,7,8,9,16,19,21,32,34,36,39,40在10-5 M浓度下,对APAP(扑热息痛)引起的肝细胞损伤有保护作用;送筛化合物在10-6 M浓度均未表现出细胞毒活性。

  以上研究丰富了四季青化学成分的研究内容,为其抗炎、保肝等活性成分的深入研究以及进一步开发利用奠定了物质基础。

  This dissertation summarizes the chemical constituents and pharmacological activities of the genus of Ilex. 

    “Ilex chinensis Folium” is the dried leaves of Ilex chinensis Sims, which belongs to the genus Ilex of Aquifoliaceae. It is collected in autumn and winter, and dried in the sun. The actions of “Ilex chinensis Folium” are to clear away heat and toxin, and reduce swelling and dissipation blood stasis. The indications are used in treating diseases like acute laryngopharyngitis, acute tracheobronchitis, dysentery, burns and skin ulcer.

    42 compounds (1-42) were isolated by various chromatographic techniques from the leaves of I. chinensis Sims, and the structures were elucidated and identified by spectroscopic analysis, including 11 new compounds (1-11). The compounds include 15 triterpenoids: ilexchinenins A, B (1, 2), ursolic acid (12), rotundic acid isopropylidene (13), 2α,3β-dihydroxy-23-norurs-4(24)-12-dien-28-oic acid (14), rotundic acid (15), 3β-hydroxy-27-p-(Z)-coumaroyloxyurs-12-en-28-oic-acid (16), 23-hydroxyursolic acid (17), 3β,23-dihydroxyurs-12,18(19)-dien-28-oic acid (18), 2α,3β,23-trihydroxyurs-12,18-dien-28-oic acid (19), asiatic acid (20), 3β-hydroxy-27-p-(Z)-coumaroyloxyolean-12-en-28-oic-acid (21), ilexosapogenin A (22), 2α,3β-dihydroxy-23-norolean-4(24)-12(13)-dien-28-oic acid (23), 29-hydroxyhederagenin (24); 19 triterpenoid saponins: ilexchinenosides A-I (3-11), peduncloside (25), ilexoside B (26), pomolic acid 28-O-β-D-glucopyranoside (27), rotungenoside (28), 3β,23-dihydroxyurs-12,18-dien-28-oic acid 28-O-β-D-glucopyranosyl ester (29), 3β,23-dihydroxyurs-12,19-dien-28-oic acid 28-O-β-D-glucopyranosyl ester (30), 3β,23-dihydroxyurs-12,19(29)-dien-28-O-β-D-

glucopyranosyl ester (31), ilexoside A (32), 28-O-β-D-glucopyranoside siaresinolic acid (33), 28-O-β-D-glucopyranoside spathodic acid (34); 4 phenolic acids: vanillic acid (35), p-hydroxy-benzoic acid (36), (E)-4-hydroxycinnamic acid (37), protocatechuic acid (38); 2 steroid compounds: β-sitosterol (39), daucosterol (40); 1 coumarin: esculetin (41); 1 fatty acid: palmitic acid (42). Compounds 19, 21, 23, 24 are isolated from the genus of Ilex for the first time. In addition to compounds 12, 15, 25, 26, 32, 38-40, othe compounds are isolated from I. chinensis Sims for the first time.

    Some isolated compounds were tested in several in vitro pharmaceutical activity assays. Compounds 1, 3, 5, 12, 16, 21, 29 and 30 exhibited significant inhibitory effects on NO production of lipopolysaccharide (LPS)-induced murine macrophages; Compounds 4, 7, 8, 9, 16, 19, 21, 32, 34, 36, 39 and 40 exhibited potent hepatoprotective activity against N-acetyl-p-aminophenol (APAP)-induced toxicity in vitro; All tested compounds showed no cytotoxic activities.

    The above researches enrich the contents of chemical constituents of “Ilex chinensis Folium” and establish good foundation for further research of the anti-antiinflammatory and other activities constituents.
